This study expands understanding of opsin 3 (OPN3) function and of skin pigmentation. The findings presented here reveal that the nonvisual OPN3 modulates the pigmentation of human epidermal melanocytes—the melanin-producing cells of the skin—by controlling the activity of the main pigmentation receptor, melanocortin 1 receptor (MC1R). Abnormalities associated with melanocytes are often caused by disruptions to or loss of function of the pigmentation and its related factors. These pigmentary disorders are classified as hyperpigmentation disorder, hypopigmentation disorder, mixed hyperpigmentation disorder and mixed hypopigmentation disorder which are further subclassified into acquired and congenital [50] . Melanocytes migrate to the uveal tract of the eye (iris, ciliary body, and choroid), the leptomeninges, and the cochlea of the inner ear, as well as to the epidermis and hair follicle. Cutaneous melanocytes can also arise from Schwann cell precursors located along nerves in the skin, which also originate from the neural crest.
